Initial studies showed that mouse erythroleukemia (MEL) cells transduced with AnkT9W exhibited far greater hemoglobin synthesis than cells transduced with a T9W vector that didn’t include the ankyrin insulator. This enhanced hemoglobin synthesis was associated with increased levels of β-globin mRNA and translation. Importantly, the ankyrin element was stably integrated in the MEL cells.

Transduced cells from patients with β+ thalassemia (i.e., that already produce some Hb A) started to produce levels of total Hb A (in other words transgenic Hb A plus endogenous Hb A) that were equivalent to those in healthy cells. Even totally Hb A-deficient β0 cells were capable of producing levels of transgenic Hb A comparable to about 55% of normal, after transduction with AnkT9W.

Overall, it was evident that β0/0 cells required higher amounts of vector to reach therapeutic levels of Hb, whereas Β0/+ and β+/+ cells already expressed some endogenous HbA and reached curative levels at lower vector copy numbers (VCNs). Perkins Volunteer Fire Department recently added a new tanker to its fleet.

Fire Chief Joe Barta said the department received the equipment at no cost from the Oklahoma Department of Forestry through a Department of Defense program.

Barta said members of the volunteer fire department provided that work during their free time over the past year.

For a number of Christians, evolution kills any possibility of a magnificent Creator who formed the cosmos from the void, the Artist who paints the horizons with wondrous sunsets, the Potter who molds the human clay, the Author who writes the story of life on Earth.

Without an author, humans can no longer be sure that the story’s end will be happy. Without a painter, humans cannot behold the beauty of God within the natural world. Without a potter, humans are little more than off-center lumps of clay twirling awkwardly on the wheel of life. Without a Creator, humans can no longer be made in his image.

I should know, because I used to be one of those Christians. For years, I thought of Darwin as an agent of devilish misinformation who stood against God’s ultimate, infallible truth when it came to creation. To me, evolution was one of the great spiritual battles of modern times, and I was a foot soldier on the side of God fighting against an onslaught of evolutionary godlessness.

For me at the time, a Christian who accepted the theory of evolution as a scientific truth wasn’t just as signal of a lack of faith or a capitulation to secular culture.

Instead, it signaled the end – the death – of the Creator God.

But such a death also signals liberation.

So in honor of Darwin’s birthday and Evolution Weekend, I am giving thanks as a Christian like others in the Clergy Letter Project for the courageous brilliance of the man who pioneered the theory of evolution and for the invigorating liberation he has offered people of faith. The Pioneers Inn got a bailout last year, but not from Uncle Sam.

People who live here and in nearby communities pulled together and raised about $3,500 to keep the town's only business open.

"We want this town to have something when we drive through," said Marjure Coordsen, sitting in a booth with friends Alvina Krueger and Kathy Schroeder on Friday morning.

The women didn't just help save a historic 125-year-old building -- made of quarried limestone blocks -- that needed extensive repairs and new equipment. They may have saved the lifeblood of the town, which has as its slogan: "There is a balm in Gilead."

People go to the Pioneers Inn for more than food. This is the place where birthdays are celebrated. It's where local clubs get together, and the men and women meet for coffee, sometimes twice a day.

During Christmas, owners Karen and Lyle Keilwitz coax Santa Claus to visit and hire a horse-drawn sleigh for the kids. They follow it up with an Easter egg hunt in the spring.

"I would have to move out of town," said Robert Schroeder, who drops by the Pioneers Inn four times a day. "I think it's the right thing to do."

So far, new studs and walls have been put in to help insulate a drafty storage room, and some customers cleaned up the basement so the sagging floor can be raised a few inches. More work is scheduled in February.

"I think it's wonderful," said Karen Keilwitz. "It's amazing who's all helping out. People in Fairbury and Hebron are donating to keep this place open. It's kind of weird."

But there may be ulterior motives at work.

Already at the helm of a 150-year-old hotel, as well as a cottage, cabin and bunkhouse, one Scots couple decided to create two more boltholes. Located on the awe-inspiring shoreline of Loch Eriboll on Scotland’s north-western tip, these would be aimed at couples seeking complete escapism, but not from their dogs.

Fiona and Robbie are locals who live in the village of Durness, six miles from the croft land (owned by Robbie’s family) where the new buildings are situated, and on which they had contemplated building their own home. “We never quite got round to that,” says Fiona.

The carbon-neutral nature of the proposed buildings (described as “crofts”, Fiona explains, only for want of a better word) won favour with local planners. Community support was also strong, and a precedent for unconventional architecture had already been set; the award-winning home of ceramic artist Lotte Glob, mounted on stilts and set beneath a curved roof of patinated copper is located nearby.

While Fiona and Robbie’s crofts, collectively dubbed “Croft 103”, both face south to maximise solar gain, they are also positioned to allow for privacy. So while Hill Croft curves in a fan shape into the hillside above Loch Eriboll, the rainbow arc of Shore Croft sits much closer to the water’s edge.

Both embrace what Fiona describes as “honest” materials, “We didn’t want anything ostentatious or twee,” she says, “These spaces had to be very sexy and sleek yet undoubtedly ‘Highland’.”

The honey-coloured stone used to clad the exterior walls, and to create drystane dykes, was sourced from the croft itself, while Douglas fir for the buildings’ frameworks was locally sourced and milled by Matt Stevenson of Carbonlite Design + Build, the Ross-shire based company that guided the couple through the project.

Traditional stone and timber nevertheless meld in these designs with sharp contemporary finishes including zinc roofs and glass walls to create buildings that are quite out of the ordinary. An abundance of south-facing glass allows each space to be heated largely by passive solar gain. However, hot water (for the highly desirable external tubs as well as those inside) comes largely from solar panels positioned remotely from the buildings. Meanwhile a wind turbine generates electricity, with its surplus energy being sold back to the grid.

Amongst other unconventional items on the couple’s shopping list were sheep’s wool (for insulation) and tyre bales (made from recycled car tyres) that make a versatile building block.

Working in such a remote location did pose challenges. Sunday was perfect weather for a day on the ice - sunny, clear and calm. Operating on "mountain time," after a leisurely breakfast, Cody and I made our way to the reservoir where we found a thick icecap that was responding noisily to the prior night's cold. Growing ice is a lot like rice cereal after adding milk; it snaps, crackles and pops.

Our first hole revealed more than 12 inches of clear, hard ice, which was plenty to bear our weight. The end-of-year holidays aren't likely to be happy ones for 200 workers in the San Francisco Bay Area who have recently been fired—or are about to be. Today, just before Christmas Eve, yet another round of workers will be cut loose from Pacific Steel Castings Foundry in Berkeley, Calif., the largest steel foundry west of the Mississippi.

The company has laid off 22 workers each Friday since October, and will continue to do so until January 4.

The firings are fallout from a silent Immigrations and Customs Enforcement raid. The federal agency started its I-9 employee audit of worker immigration status back in February, as David Bacon reported for In These Times. The I-9 employee audits target employers who hire workers who are undocumented. The employers can face fines, jail time and the loss of government contracts.

ICE claims the 200 workers, many of whom had been employed at Pacific Steel for decades, had no legal immigration status. Employers notified of an audit have to turn over I-9 documents, which include Social Scurity numbers, birth dates and statements by workers about their citizenship status.

A spokeswoman for ICE told KTVU television station that the agency usually acts on tips when deciding which businesses to audit.

Pacific Steel, the fourth largest foundry in the United States, says it began the firing of more than a third of its workforce after pressure by federal immigration authorities. The company makes steel parts for trucks and custom molds with revenue totaling $100 million a year.

Pacific Steel spokeswoman Elisabeth Jewel told the Contra Costa Times, "This was not an action that the company invited. It's been a very wrenching experience for the company to lose so many highly valued, highly skilled employees, some of whom have been there for decades."

The workers’ union, government officials and community members appealed the audit and mass firings. The only thing it did was delay the firings of the workers the company identified as “nonresidents” until October.
